چکیده

With the use of photoelectric scanning optics on the analytical ultracentrifuge molecular weights on human plasminogen and plasmin have been determined at levels as low as 50 pg per ml and as low as 10 pg per ml for the sedimentation coefficient. The results give a molecular weight of 81,000 for human plasminogen and 75,400 for human plasmin. The molecular weight difference has been verified with differential sedimentation equilibrium techniques. The $0, 1u value of human plasminogen and plasmin were determined to be 4.2 S and 3.9 S, respectively.

Preparation of Plasminogen by Affinity Chromatography

Background: Plasminogen is one of the compounds derived from human plasma. Activation of plasminogen produces plasmin. Plasmin is able to lyse fibrinogen, fibrin, and some other human plasma proteins. The aim of the present work was to study the separation of human plasminogen by affinity chromatography using gel lysine Sepharose. Materials and Methods: Normal human plasma was used as the...

اثر آنتی بادی های منوکلونال ضد پلاسمینوژن انسانی بر فعال شدن گلو-پلاسمینوژن انسانی بوسیله فعال کننده های پلاسمینوژن

Background: Human plasminogen is a plasma glycoprotein synthesized mainly in the liver. Conversion of plasminogen to plasmin by plasminogen activators is a key event in the fibrinolytic system. In this study, we investigated the effects of two anti-human plasminogen monoclonal antibodies, A1D12 and MC2B8 on Glu-plasminogen activation in presence of u-PA, t-PA and streptokinase. Methods: Produci...

On the formation of different types of plasmin by streptokinase activation.

Plasminogen, a normal constituent of human blood plasma, can be activated in various ways to yield the proteolytic enzyme plasmin capable of dissolving fibrin clots and of hydrolyzing a variety of other proteins and synthetic amino acid esters. The activation by the bacterial product streptokinase has been widely studied in the past and appears to proceed through a two-stage reaction: the inter...

Fibrinolysis--a review.

The function of fibrinolysis is to dissolve fibrin clots. The agent of fibrinolysis is plasmin, a glycoprotein with gram molecular weight (GMW) of 90,000. Under natural conditions, plasminogen is converted to plasmin by tissue plasminogen activator (TPA). Activation occurs on the fibrin surface, thus confining proteolytic activity to the appropriate site. Tissue plasminogen activator, produced ...

Inhibition of plasminogen activator inhibitor-1 activity by two diketopiperazines, XR330 and XR334 produced by Streptomyces sp.

Two diketopiperazines, XR334 (1) and the novel compound XR330 (2), were isolated from the lyophilised biomass of an unidentified Streptomyces sp. Their structures were elucidated on the basis of spectroscopic studies and confirmed by chemical synthesis. Both compounds inhibited plasminogen activator inhibitor-1 activity in an amidolytic assay of tissue plasminogen activator mediated plasmin gen...
